Growth , Photosynthetic Rate and Stomatal Function of Spathiphyllum In Vitro as Affected by PPF and NAEH


Abstract
The shoot of spathiphyllum (Spathiphyllum Schott cvs. Merry and Cupido) was cultured on the vessel containing Murashige and Skoog agar medium with no added sugar, but supplemented with 1.25 meq¡¤L^(-1) H©üPO©þ^- . Treatments used were two levels each of photosynthetic photon flux (PPF) (70 and 150 ¥ìmol¡¤m^(-2)¡¤s^(-1)) and number of air exchanges per hour (NAEH) (0.1, no gas permeable membrane filter, and 2.8 h^(-1), one gas permeable membrane filter) for in vitro cultures. Four explants were planted in each 3.7 ¡¿ 10^(-4) §© GA_7 Magenta box containing 1.5 ¡¿ 10^(-5) §© medium. The pH of the medium was adjusted to 5.70 before autoclaving. Explants were cultured under cool white fluorescent lamps with the supplement of 1,000 ¥ì§ß¡¤§ß^(-1) CO©ü during the photoperiod, and at an air temperature of 25¡¾1¡É and relative humidity of 70-80%. In treatment of 2.8 h^(-1) NAEH, a 10 §® round hole made on the vessel cap was sealed with a microporous filter. Growth responses of the two cultivars were similar. Both root length and leaf area of spathiphyllum ¢¥Merry¢¥ were significantly enhanced by 70 ¥ì§ß¡¤m^(-2)¡¤s^(-1) PPF. Chlorophyll content was suppressed by high PPF, and increased under high NAEH. Spathiphyllum ¢¥Cupido¢¥ cultured under 2.8 h^(-1) NAEH showed an increased net photosynthetic rate and stomatal conductance. Leaves produced under high NAEH had greater number of stomata than those produced under low NAEH condition. Leaves of spathiphyllum plantlets cultured under 0.1 h^(-1) NAEH had closed stomata, whereas those cultured under 2.8 h^(-1) NAEH had open ones.
